Considering NFL teams have 48 players (or 53, a bit confused) active roaster, they can still manage it.
The rosters are very big, so they should be able to weather it. However, teams usually sign free agents to fill specific needs and positions are specialized enough that it takes about 45 guys just to have one backup at each position.
